ENB-812 is a preclinical, first-in-class precision oncology **small molecule** inhibitor of protein kinase, membrane associated tyrosine/threonine 1 (PKMYT1), being developed by Engine Biosciences for biomarker-defined subsets of solid tumors including ovarian, liver, colorectal, prostate, and lung cancers.[4][9][10][12] Engine independently identified PKMYT1 as a synthetic lethal target and used its network biology and machine learning platforms to discover ENB-812 and associated patient selection biomarkers, generating preclinical proof-of-concept across multiple tumor models and advancing the program into IND-enabling studies.[1][9][10][12] By inhibiting PKMYT1, a cell-cycle regulatory kinase, ENB-812 is intended to exploit synthetic lethal vulnerabilities in tumors with specific genetic alterations, enabling selective antitumor activity while sparing normal tissues.[1][4][9][10]
