ENB101-LNP is an investigational RNA interference (RNAi) therapeutic developed by EnhancedBio Inc. It consists of ionizable lipid nanoparticles (LNPs) encapsulating small interfering RNA (siRNA) specifically designed to target the E6 and E7 oncoproteins of human papillomavirus type 16 (HPV16). These oncoproteins are critical for the maintenance of the malignant phenotype in HPV-associated cancers, such as cervical cancer, by degrading tumor suppressors like p53 and pRb. By silencing E6 and E7 expression, ENB101-LNP restores p53 and p21 levels, inducing apoptosis and cell cycle arrest. Preclinical data presented at AACR 2023 demonstrated significant anti-tumor efficacy in CaSki cell xenograft models, particularly when combined with cisplatin, showing enhanced tumor growth inhibition and reduction of PD-L1 expression.
