Overview
Encenicline is a selective, orally active small molecule that acts as a partial agonist of the alpha 7 nicotinic acetylcholine receptor (α7-nAChR). It was developed for the treatment of cognitive impairment associated with schizophrenia and Alzheimer's disease. The drug enhances cholinergic neurotransmission by selectively targeting α7-nAChRs, which are implicated in cognitive processes. Encenicline penetrates the blood-brain barrier and has been shown in preclinical studies to improve memory performance by potentiating acetylcholine responses. Clinical trials reached phase III but were discontinued due to lack of efficacy and concerns about gastrointestinal side effects at higher doses. The main developers were FORUM Pharmaceuticals and Mitsubishi Tanabe Pharma[1][2][3].
