Overview

Encofosbuvir (HEC110114) is a small-molecule nucleotide analog prodrug developed by Sunshine Lake Pharma for the treatment of chronic hepatitis C virus (HCV) infection. As a direct-acting antiviral (DAA), it targets the NS5B RNA-dependent RNA polymerase. Once metabolized into its active triphosphate form, it acts as a defective substrate for the NS5B polymerase, leading to premature viral RNA chain termination. Encofosbuvir demonstrates pan-genotypic activity across HCV genotypes 1 through 6. It was approved in China in March 2025 as part of a domestically produced pan-genotypic regimen, typically co-administered with the NS5A inhibitor netanasvir.
