Overview

Encorafenib is a small molecule kinase inhibitor used as an antineoplastic agent. It selectively inhibits the BRAF serine/threonine kinase, particularly in tumors with activating BRAF V600E or V600K mutations. By blocking mutant BRAF, encorafenib disrupts the MAPK/ERK signaling pathway, leading to decreased cell proliferation and tumor growth. It is primarily indicated for use in combination with other agents (binimetinib or cetuximab) for unresectable or metastatic melanoma, metastatic colorectal cancer (CRC), and non-small cell lung cancer (NSCLC) harboring specific BRAF mutations. Encorafenib was developed by Array BioPharma and Novartis[3][5].
