Overview

Encorafenib, binimetinib, and cetuximab is a three-drug combination regimen used in oncology. Encorafenib is a small molecule inhibitor of the BRAF kinase (including mutant BRAF V600E), while binimetinib is a small molecule inhibitor of MEK1/2 kinases. Cetuximab is a monoclonal antibody targeting the epidermal growth factor receptor (EGFR). This triplet combination targets multiple nodes in the RAS/RAF/MEK/ERK signaling pathway, which is frequently dysregulated in cancers with BRAF V600E mutations. The regimen has been studied primarily for metastatic colorectal cancer with BRAF V600E mutation and unresectable or metastatic melanoma with BRAF V600 mutation[3][5][8]. The BEACON CRC phase III trial established this triplet as an option for previously treated patients with metastatic colorectal cancer harboring the BRAF V600E mutation[8]. Encorafenib and binimetinib are developed by Pfizer; cetuximab was developed by ImClone Systems (now part of Eli Lilly) and marketed by Merck KGaA outside North America.
