Overview

PER-001 (endonentan) is a novel, first-in-class small molecule endothelin A receptor antagonist developed as a biodegradable intravitreal implant for the treatment of back-of-the-eye diseases. Endothelin is the most potent vasoconstrictor in the human body and plays a role in inflammation and cell death through its receptors expressed in retinal vascular and neuroretinal cells. By antagonizing the endothelin A receptor, PER-001 aims to improve optic nerve head blood flow, provide neuroprotection, and address ischemia-driven pathologies independent of intraocular pressure. Clinical data from Phase 1/2a trials show that PER-001 was well-tolerated with promising improvements in visual field mean deviation and retinal nerve fiber layer thickness for patients with glaucoma. The drug is administered via a bio-erodible PLGA polymer matrix implant designed for sustained release every six months[1][3][6][7].
