A combination regimen consisting of **EndoTAG-1**, **paclitaxel**, and **gemcitabine** investigated primarily for advanced or metastatic cancers such as pancreatic cancer. **EndoTAG-1** is a cationic liposomal formulation of paclitaxel designed for targeted delivery to tumor vasculature. **Paclitaxel** is a microtubule-stabilizing agent that disrupts cell division, while **gemcitabine** is a nucleoside analog that inhibits DNA synthesis. The combination leverages different antineoplastic mechanisms: targeted delivery to tumor vessels with EndoTAG-1, cell cycle arrest with paclitaxel, and inhibition of DNA replication with gemcitabine. The regimen has been evaluated in clinical trials, mainly for pancreatic cancer, with evidence of improved efficacy compared to gemcitabine alone.
