Overview

ENG-E2 is a combination drug consisting of etonogestrel (a progestin) and 17β-estradiol (a natural estrogen), formulated as a contraceptive vaginal ring. The mechanism of action involves suppression of ovulation through inhibition of gonadotropin secretion by the pituitary gland, primarily via negative feedback on the hypothalamic-pituitary-gonadal axis. Etonogestrel acts as a progestin to inhibit ovulation and thicken cervical mucus, while estradiol provides estrogenic activity to stabilize the endometrium and reduce breakthrough bleeding. The product was developed for use in women of reproductive age for contraception[1][6][10].
