Overview
Engedi1000 (E1K) is a synthetic peptide drug developed for the treatment of osteoarthritis. It works by modulating the signaling pathway of transforming growth factor beta 1 (TGF-β1), which is implicated in cartilage degeneration. Specifically, Engedi1000 binds to TGF-β1 and selectively blocks the Smad1/5/8 pathway that promotes cartilage tissue degeneration while maintaining the Smad2/3 pathway that supports cartilage regeneration. Additionally, it reduces nerve growth factor (NGF) gene expression to provide pain relief. The drug has shown efficacy in preclinical models and has completed phase II clinical trials for knee osteoarthritis in South Korea[1][2][4][5].
