Engensis is a non-viral gene therapy developed by Helixmith that uses plasmid DNA to deliver the human hepatocyte growth factor (HGF) gene directly into muscle tissue. The plasmid encodes two natural isoforms of HGF, which together promote angiogenesis (formation of new blood vessels), nerve regeneration, and muscle repair. Engensis is administered via intramuscular injection and does not integrate into the patient's genome; instead, it provides transient expression of HGF protein requiring repeated dosing. It is being investigated for several indications including amyotrophic lateral sclerosis (ALS), painful diabetic neuropathy, critical limb ischemia, Charcot-Marie-Tooth disease type 1A (CMT1A), peripheral artery disease, and diabetic foot ulcers. Its mechanism centers on boosting local HGF production to support tissue regeneration and neuroprotection.
