engEx-AAV is an exosome-based gene therapy delivery platform originally developed by Codiak BioSciences and acquired by Evox Therapeutics in 2023. The technology involves the encapsulation of adeno-associated virus (AAV) vectors within engineered exosomes to enhance delivery efficiency and evade neutralizing antibodies. It utilizes a proprietary scaffold based on the BASP1 protein and a camelid nanobody (VHH) to facilitate the loading of AAV capsids into the exosomal lumen. This approach is designed to enable targeted delivery to specific tissues, such as the retina for ocular diseases, while reducing systemic exposure and improving the breadth of gene expression compared to standard AAV vectors.
