Overview

The engineered anti-EGFR IgA antibody is a research-stage biologic (monoclonal antibody of the IgA isotype) developed and studied by Radboud Universitair Medisch Centrum (Radboud UMC) in the Netherlands. It is engineered to target the Epidermal Growth Factor Receptor (EGFR), a receptor tyrosine kinase overexpressed in many cancers, including carcinomas. Unlike conventional IgG-based therapeutic antibodies (such as cetuximab), this antibody is of the IgA class, designed to engage the Immunoglobulin alpha Fc receptor (FcαRI/CD89) on myeloid effector cells such as neutrophils and monocytes/macrophages. This engagement recruits innate immune cells to kill tumor cells via mechanisms such as antibody-dependent cellular cytotoxicity (ADCC) and phagocytosis. It is being evaluated in ex vivo translational studies (such as the INVOLVE study) using freshly donated blood from healthy adult volunteers to isolate myeloid effector cells and test the antibody's ability to mediate tumor cell killing and phagocytosis in laboratory assays.
