Overview

Engineered Clostridium butyricum GLP-1 is an orally administered live biotherapeutic product (LBP) consisting of a genetically modified strain of the probiotic bacterium *Clostridium butyricum* engineered to express and secrete glucagon-like peptide-1 (GLP-1). Developed by the Reproductive Hospital Affiliated to Jiangxi University of Traditional Chinese Medicine, this therapeutic approach leverages the gut-colonizing capabilities of *C. butyricum* to provide a continuous, localized delivery of GLP-1 within the gastrointestinal tract. The secreted GLP-1 acts as an agonist at the glucagon-like peptide 1 receptor (GLP1R), which promotes satiety, slows gastric emptying, and improves insulin sensitivity. This modality aims to overcome the limitations of injectable GLP-1 receptor agonists by offering a non-invasive, oral delivery system that potentially modulates the gut microbiome while providing metabolic benefits. It is currently under investigation in clinical trials (e.g., ChiCTR2500104877) for the treatment of obesity.
