Overview

**Engineered E. coli co-displaying IL-12 + ADA** is a preclinical live microbial cancer immunotherapy developed at Dana-Farber Cancer Institute. The product uses non-pathogenic *Escherichia coli* K-12 DH5α engineered with outer-membrane display scaffolds to localize surface-tethered interleukin-12 and adenosine deaminase in tumors. Surface IL-12 activates NK cells and T cells, while ADA depletes immunosuppressive adenosine by converting it to inosine. In mouse acute myeloid leukemia and melanoma models, the dual-function bacterial construct produced tumor control, enhanced NK-cell cytotoxicity, and prolonged survival; intratumoral administration produced complete regression in a B16-F10 melanoma model. The reported program remains preclinical and does not have an assigned INN, sponsor development code, or marketed brand name.
