Drug intelligence / Profile preview
Engineered MyD88 fusion proteins are a class of synthetic proteins designed to enhance the fitness, expansion, and persistence of chimeric antigen receptor (CAR) T-cell therapies. MyD88 is an essential scaffold protein in the innate immune system that propagates pro-inflammatory signals. While early chemically induced dimerization (CID) platforms fusing MyD88 to the intracellular domain of CD40 (such as the inducible MyD88/CD40 or iMC switch) successfully boosted CAR-T cell activity, they were associated with clinical toxicity due to excessive systemic cytokine release. To address this, next-generation engineered MyD88 fusion proteins are designed with modulated assembly thresholds (by altering the stability of the dimeric seed) and tuned signaling motifs (such as natural or synthetic TRAF recruitment motifs) to achieve a safer, more controlled pro-inflammatory profile.
