Englumafusp alfa is an investigational antibody-like fusion protein designed to target both CD19 on B cells and Tumor necrosis factor receptor superfamily member 9 (4‑1BB/CD137) on T cells and other immune effector cells. Its mechanism of action involves simultaneous binding to CD19 and 4-1BB, forming a trimeric complex that co-stimulates T cells via Tumor necrosis factor receptor superfamily member 9 (4‑1BB/CD137) agonism in the presence of a T-cell receptor signal and strictly dependent on CD19 crosslinking. This dual targeting aims to enhance anti-tumor immune responses by boosting T-cell effector functions while preventing T-cell anergy. Englumafusp alfa is being developed primarily for the treatment of relapsed or refractory B-cell non-Hodgkin lymphoma (NHL), including diffuse large B-cell lymphoma (DLBCL). It is currently under investigation in phase I/II clinical trials as part of combination regimens with glofitamab (an anti-CD20xCD3 bispecific antibody) and obinutuzumab.
