EngTreg is an engineered regulatory T cell (Treg) therapy platform developed by GentiBio for the treatment of autoimmune and inflammatory diseases. The platform utilizes a gene-editing approach (dual AAV or nonviral) to convert bulk CD4+ T cells into stable, functional Tregs by ensuring the constitutive expression of the master transcription factor FOXP3. To address the challenge of inconsistent growth factor availability in the body, EngTregs are engineered with a chemically inducible signaling complex that provides selective IL-2 signaling upon the administration of rapamycin, allowing for the enrichment and survival of the edited cells. The platform is modular and can be tailored for specific indications: TCR-EngTregs (targeting IGRP for Type 1 Diabetes), Gut-targeted EngTregs (for Inflammatory Bowel Disease), CAR19 EngTregs (for B-cell driven autoimmunity), and Tissue EngTregs (expressing the alarmin receptor ST2 for acute tissue repair). Additionally, EngTregs incorporate hypoimmune engineering to prevent rejection by T, B, and NK cells, enabling an allogeneic, off-the-shelf therapeutic strategy.
