Enhanced Autologous TCR-T is a preclinical-stage cell therapy program developed by Lion TCR. This approach utilizes autologous T-cell receptor (TCR) engineered T cells that are further modified using antisense oligonucleotides (ASOs) to modulate gene expression. The goal of this dual approach is to enhance the antiviral activity, fitness, and persistence of the T cells when targeting viral antigen-expressing cells, such as those found in chronic hepatitis B virus (HBV) infections or HBV-associated hepatocellular carcinoma. By incorporating ASOs, the therapy can potentially downregulate inhibitory pathways or resistance mechanisms within the T cells or the surrounding microenvironment, providing a more robust immune response compared to standard TCR-T therapies. This platform is distinct from Lion TCR's lead LioCyx-M platform, which primarily utilizes mRNA transfection for TCR expression.
