EnkaStim + Keytruda is a combination immunotherapy regimen being developed by Alphageneron for the treatment of metastatic Stage IV non-small cell lung cancer (NSCLC). The combination consists of EnkaStim, an autologous cell therapy comprising non-expanded natural killer (NK) cells targeted against membrane-bound Heat Shock Protein 70 (mHsp70), and Keytruda (pembrolizumab), a programmed cell death protein 1 (PD-1) blocking monoclonal antibody. mHsp70 is a tumor-specific antigen frequently expressed on the surface of various cancer cells, including NSCLC, but absent on normal tissues. By combining mHsp70-targeted NK cell cytotoxicity with the checkpoint inhibition provided by pembrolizumab, the therapy aims to leverage the direct cytotoxic activity of targeted NK cells while simultaneously reversing T-cell exhaustion to enhance the overall anti-tumor immune response. The program is currently in Phase 1 clinical development.
