Overview

**Enlicitide + digoxin** is a combination of two pharmaceutical agents: **enlicitide**, a novel oral macrocyclic peptide, and **digoxin**, a cardiac glycoside. This combination is currently under investigation primarily for pharmacokinetic interaction purposes, specifically to assess the effect of enlicitide on the disposition of digoxin in healthy adults[1]. Enlicitide itself is in advanced clinical development (Phase 3) as a daily oral proprotein convertase subtilisin/kexin type 9 (PCSK9) inhibitor for hypercholesterolemia and related lipid disorders[2][3][4][5][6]. Its mechanism involves binding and inhibiting PCSK9, thereby increasing LDL receptor availability and lowering LDL cholesterol levels. Digoxin is a longstanding treatment for heart failure and certain arrhythmias, acting principally by inhibiting sodium/potassium-ATPase, which increases cardiac contractility. No clinical indication is currently established for the combination—its use in trials centers on pharmacokinetic assessments[1].
