Overview

Enmetazobactam is a novel penicillanic acid sulfone beta-lactamase inhibitor structurally related to tazobactam. It is used in combination with cefepime, a fourth-generation cephalosporin, for the treatment of complicated urinary tract infections (cUTI), including pyelonephritis, in adults. Enmetazobactam works by inhibiting extended-spectrum beta-lactamases (ESBLs) and other class A beta-lactamases such as CTX-M, TEM, and SHV enzymes that confer resistance to many antibiotics. By blocking these enzymes, enmetazobactam protects cefepime from degradation and restores its antibacterial activity against ESBL-producing Enterobacterales and other resistant gram-negative bacteria. The combination has demonstrated superior efficacy compared to piperacillin/tazobactam in clinical trials and offers a carbapenem-sparing option for serious infections caused by multidrug-resistant organisms[2][5][7].
