ENO1 shRNA is a research-stage gene-silencing construct designed to knock down the expression of Enolase 1 (ENO1), also known as alpha-enolase. ENO1 is a key glycolytic enzyme that is frequently overexpressed in various malignancies, including acute myeloid leukemia (AML), gastric cancer, and pancreatic ductal adenocarcinoma, where it promotes tumor cell proliferation, migration, and chemoresistance. By delivering short hairpin RNA sequences via lentiviral vectors or plasmids, this tool triggers the RNA interference (RNAi) pathway to degrade ENO1 mRNA, thereby reducing protein levels. Preclinical studies have demonstrated that ENO1 knockdown can inhibit the MAPK/ERK signaling pathway, induce apoptosis, and sensitize cancer cells to agents like cisplatin and gemcitabine. While commercially available as a laboratory reagent from suppliers such as OriGene, it is currently utilized primarily in academic research and preclinical validation of ENO1 as a therapeutic target.
