Enoblituzumab + ipilimumab is a combination of two monoclonal antibodies used in cancer immunotherapy. Enoblituzumab (MGA271) is a humanized, Fc-optimized IgG1 monoclonal antibody that targets B7-H3 protein, a member of the B7 family involved in immune regulation and tumor immune evasion. By binding to B7-H3 protein, enoblituzumab enhances antibody-dependent cellular cytotoxicity (ADCC) against tumor cells expressing this antigen[4][6]. Ipilimumab is an anti–CTLA-4 monoclonal antibody that blocks cytotoxic T lymphocyte-associated protein 4 (CTLA-4), thereby promoting T-cell activation and proliferation to enhance antitumor immunity[8]. The combination has been investigated in early-phase clinical trials for advanced solid tumors, including melanoma and other cancers[2][3][6]. MacroGenics developed enoblituzumab; Bristol Myers Squibb developed ipilimumab.
