Overview

The **eNOS plasmid** is a plasmid DNA construct encoding the human endothelial nitric oxide synthase (eNOS) gene. It is used as a gene therapy tool for the local or systemic delivery of eNOS to vascular tissues, primarily through non-viral methods, such as stent-based delivery or direct tissue injection. Mechanistically, the plasmid promotes the expression of eNOS, an enzyme that synthesizes nitric oxide (NO), a critical signaling molecule involved in vascular homeostasis, vasodilation, and inhibition of smooth muscle proliferation. By elevating local NO levels, eNOS plasmid delivery reduces neointimal hyperplasia, promotes re-endothelialization, and has shown efficacy in preclinical models for conditions such as restenosis, essential hypertension, and pulmonary hypertension. eNOS gene therapy can be administered as naked plasmid DNA, or in combination with delivery systems such as lipopolyplexes or minicircle DNA, to enhance cellular uptake and transgene expression. The therapeutic strategy aims to restore or augment endothelial function and vascular repair through the biological actions of NO[1][3][4].
