Enoticumab (REGN421) is a fully human IgG1 monoclonal antibody that targets delta-like protein 4 (DLL4), a ligand in the Notch signaling pathway. By binding to DLL4, enoticumab disrupts Notch-mediated signaling, which plays a critical role in angiogenesis—the formation of new blood vessels—particularly within tumors. Inhibition of DLL4-Notch interaction leads to the development of nonfunctional tumor vasculature and reduced tumor growth. Enoticumab has been investigated primarily for the treatment of advanced solid tumors and cancers, with preclinical studies showing antitumor effects through both anti-angiogenic mechanisms and reduction of cancer stem cells. The drug was developed by Regeneron Pharmaceuticals in collaboration with Sanofi[1][2][3][5].
