Enpatoran is an orally bioavailable small molecule that acts as a dual antagonist of toll-like receptor 7 and toll-like receptor 8 (TLR7/8). By inhibiting these receptors, enpatoran blocks TLR7/8-mediated pathways, which are involved in the activation of the innate immune system and production of pro-inflammatory cytokines. This mechanism provides anti-inflammatory and immunomodulatory effects, making it a potential therapy for autoimmune diseases such as systemic lupus erythematosus (SLE), cutaneous lupus erythematosus (CLE), dermatomyositis, polymyositis, and for hyperinflammatory conditions like COVID-19 pneumonia. Enpatoran has been investigated in multiple phase I and II clinical trials for these indications. The drug was developed by Merck KGaA/EMD Serono[1][2][4][5][6].
