ENPP-1-IN-1 is a small molecule inhibitor of Ectonucleotide Pyrophosphatase/Phosphodiesterase 1 (ENPP1), also referred to as NPPS in some research contexts. It is primarily utilized as a research tool to investigate the metabolic and oncogenic roles of ENPP1. Recent studies have identified ENPP1 as a critical vulnerability in RAS-mutant cancers, where it promotes hyperglycolysis by enhancing its interaction with hexokinase 1 (HK1). By inhibiting the NPPS-HK1 axis, ENPP-1-IN-1 suppresses the growth and survival of cancer cells harboring various RAS mutations in both in vitro and in vivo models. This mechanism suggests a potential therapeutic approach for targeting pan-mutant-RAS pathways through metabolic modulation.
