Ensifentrine + tiotropium is a combination therapy used in the management of chronic obstructive pulmonary disease (COPD). Ensifentrine is a novel inhaled dual inhibitor of phosphodiesterase 3 and 4 (PDE3 and PDE4), acting as both a bronchodilator and anti-inflammatory agent. It works by blocking enzymes that contribute to airway inflammation and muscle constriction, leading to relaxation and opening of air passages. Tiotropium is a long-acting muscarinic antagonist (LAMA) bronchodilator that prevents bronchoconstriction by inhibiting muscarinic receptors in the airways. The combination has been shown in clinical trials to produce additive improvements in lung function for patients with moderate-to-severe COPD who remain symptomatic despite maintenance therapy with tiotropium alone[5][6][7][8]. Ensifentrine was developed by Verona Pharma; tiotropium is marketed as Spiriva Respimat by Boehringer Ingelheim[5].
