ensovibep

Ensovibep is a first-in-class, multispecific antiviral therapeutic candidate for COVID-19. It is based on the DARPin (Designed Ankyrin Repeat Protein) platform and consists of five DARPin domains—three of which specifically bind to the receptor-binding domain (RBD) of the SARS-CoV-2 spike protein, and two that bind to human serum albumin to extend half-life. This tri-specific design enables cooperative binding to multiple sites on the viral spike protein, resulting in potent neutralization across all major variants of concern, including Omicron. Ensovibep was developed by Molecular Partners and later partnered with Novartis for late-stage development and commercialization. In clinical trials (notably EMPATHY), it demonstrated an approximately 80% reduction in risk of hospitalization or death compared to placebo in non-hospitalized patients with COVID-19[2][4][5]. The drug has been shown to be safe and well-tolerated in studies but has not yet received regulatory approval as additional data have been requested by authorities[6].
