ENT-03S is a novel, centrally acting mammalian aminosterol and a small molecule drug that functions as an inhibitor of protein tyrosine phosphatase 1B (PTP1B). It is being developed for the treatment of obesity and type 2 diabetes, with additional research interest in Alzheimer's disease. The drug acts primarily on brain circuits that regulate energy balance and metabolism by inhibiting PTP1B in the hypothalamus, thereby reversing central insulin resistance. Preclinical studies have shown that ENT-03S rapidly lowers blood glucose levels, reduces food intake and adiposity, eliminates liver fat, improves liver function, and increases insulin sensitivity. Unlike GLP-1 agonists such as semaglutide, ENT-03S produces sustained weight loss without rebound after cessation of therapy. Phase 1 clinical trials are ongoing to evaluate its safety and efficacy in obese individuals with or without type 2 diabetes[4][5][6][7][8].
