Overview

Enterolactone is a dibenzylbutyrolactone lignan and a key gut microbiota-derived metabolite produced from dietary precursors such as secoisolariciresinol and matairesinol found in plant-based foods. Recognized as a prominent phytoestrogen, it exhibits a range of biological activities including antioxidant, anti-inflammatory, and anti-proliferative effects. Research, including network pharmacology and clinical cohort analyses, suggests that enterolactone may protect against Cardiovascular-Kidney-Metabolic (CKM) syndrome by modulating glucose homeostasis, suppressing metabolic inflammation, and preserving vascular function. Its biological effects are mediated through several pathways, including the activation of estrogen signaling, inhibition of mitogen-activated protein kinase (MAPK) pathways, and suppression of the receptor for advanced glycation end products (RAGE).
