Overview

Entolimod is a recombinant protein derived from bacterial flagellin that acts as an agonist of toll-like receptor 5 (TLR5), an innate immunity receptor. By binding to and activating TLR5, entolimod triggers NF-κB signaling and mobilizes the innate immune response, leading to the expression of genes involved in anti-apoptosis, scavenging reactive oxygen species, and promoting tissue regeneration. It has demonstrated radioprotective effects in animal models by mitigating damage to hematopoietic and gastrointestinal tissues caused by radiation or chemotherapy. In oncology settings, entolimod suppresses tumor growth through stimulation of antitumor immune responses without protecting tumors from therapy. The drug is being developed for use as a radiation countermeasure under the FDA’s animal efficacy rule and as a cancer treatment under traditional approval pathways[1][3][5][6].
