Entospletinib + corticosteroids is an experimental combination therapy composed of entospletinib, a highly selective small molecule inhibitor of spleen tyrosine kinase (SYK), and systemic corticosteroids, used in clinical trials for hematological diseases. Entospletinib inhibits SYK, a key protein in B-cell receptor signaling and immune cell activation, with potential to modulate immune dysregulation implicated in diseases like chronic graft-versus-host disease (cGVHD) and acute lymphoblastic leukemia (ALL). Systemic corticosteroids are broad immunosuppressants commonly used as standard therapy in cGVHD and other autoimmune/hematological settings. The combination aims to improve response rates in conditions where corticosteroid monotherapy is insufficient, specifically targeting B-cell-mediated disease mechanisms. The combination has been explored in phase 2 clinical studies as first-line therapy for cGVHD[2][3], as well as in preclinical and early clinical studies in acute leukemia (notably KMT2A-rearranged ALL), where corticosteroids like dexamethasone have been administered alongside entospletinib[4][5].
