Entospletinib + corticosteroids is an experimental combination therapy composed of entospletinib, a highly selective small molecule inhibitor of spleen tyrosine kinase (SYK), and systemic corticosteroids, used in clinical trials for hematological diseases. Entospletinib inhibits SYK, a key protein in B-cell receptor signaling and immune cell activation, with potential to modulate immune dysregulation implicated in diseases like chronic graft-versus-host disease (cGVHD) and acute lymphoblastic leukemia (ALL). Systemic corticosteroids are broad immunosuppressants commonly used as standard therapy in cGVHD and other autoimmune/hematological settings. The combination aims to improve response rates in conditions where corticosteroid monotherapy is insufficient, specifically targeting B-cell-mediated disease mechanisms.
