Overview

**ENV-201** is an orally available small molecule inhibitor of ULK1/2 kinases, critical enzymes in the autophagy pathway, a cellular recycling process often exploited by tumor cells for nutrient supply under stress conditions like nutrient deprivation. Developed by Endeavor Biomedicines through licensing from Sanford Burnham Prebys and Salk Institute, it demonstrates single-agent preclinical activity against KRAS- and STK11-mutated cancers, where autophagy upregulation contributes to drug resistance. Currently in early-stage development as a precision oncology candidate targeting autophagy-dependent tumors.[1][5][7][9]
