Overview

Envafolimab (KN035) is a humanized monoclonal antibody that targets programmed cell death ligand 1 (PD-L1, also known as CD274). It is the first subcutaneously injectable PD-L1 inhibitor, designed for rapid administration and improved patient convenience. Envafolimab blocks the interaction between PD-L1 on tumor cells and PD-1 on T-cells, thereby enhancing T-cell mediated immune responses against cancer cells. The drug has been developed for use in various solid tumors and has shown activity in cancers with high microsatellite instability or mismatch repair deficiency. When combined with chemotherapeutic agents such as gemcitabine and oxaliplatin—both small molecule cytotoxic drugs commonly used in gastrointestinal cancers—the regimen aims to leverage both direct cytotoxicity from chemotherapy and immune-mediated anti-tumor effects from checkpoint inhibition[1][2][5][7].
