Drug intelligence / Profile preview
Enweida + nab-paclitaxel + platinum is an investigational chemo-immunotherapy combination regimen consisting of envafolimab (Enweida), a subcutaneously administered anti-PD-L1 single-domain antibody (nanobody) fused with human IgG1 Fc, combined with nanoparticle albumin-bound paclitaxel (nab-paclitaxel) and a platinum-based chemotherapeutic agent (such as carboplatin or cisplatin). This combination is primarily being evaluated as a first-line treatment for extensive-stage small cell lung cancer (ES-SCLC) and other advanced solid tumors, including squamous non-small cell lung cancer (sq-NSCLC), head and neck squamous cell carcinoma (HNSCC), and esophageal cancer. Mechanistically, envafolimab blocks the interaction between PD-1 and PD-L1 to restore T-cell-mediated anti-tumor immune responses. Concurrently, nab-paclitaxel stabilizes microtubules to arrest cell division, and the platinum agent induces DNA cross-linking to trigger apoptosis. This multi-pronged approach aims to synergize the cytotoxic effects of chemotherapy with the immune-activating properties of checkpoint inhibition.
