ENZ-8P4 is a nanoparticle-based drug delivery system consisting of the second-generation androgen receptor (AR) antagonist enzalutamide encapsulated within a biocompatible phenylpropyl polymer (8P4). This formulation was developed to address the limitations of conventional enzalutamide therapy, such as poor bioavailability and the rapid onset of drug resistance in patients with castration-resistant prostate cancer (CRPC). The 8P4 nanoparticle carrier is characterized by a small particle size and high drug-carrying capacity, which facilitates preferential accumulation in tumor tissues via the enhanced permeability and retention (EPR) effect. Preclinical studies have demonstrated that ENZ-8P4 significantly improves drug delivery efficiency, enhances cytotoxicity, and reduces the half-inhibitory concentration (IC50) compared to free enzalutamide, leading to significant inhibition of tumor growth in vivo.
