Overview

Enzalutamide is a nonsteroidal antiandrogen (NSAA) used primarily for the treatment of prostate cancer, including castration-resistant prostate cancer and metastatic castration-sensitive prostate cancer. It acts as a selective silent antagonist of the androgen receptor (AR), blocking multiple steps in the AR signaling pathway. Unlike first-generation NSAAs, enzalutamide prevents AR nuclear translocation, DNA binding, and coactivator recruitment, resulting in more effective inhibition of androgen-driven tumor growth. The drug was developed by Pfizer and Astellas, who jointly commercialize it under the brand name Xtandi. Enzalutamide is administered orally.
