Enzalutamide + paclitaxel is an investigational combination therapy consisting of enzalutamide, an androgen receptor (AR) inhibitor, and paclitaxel, a microtubule-stabilizing chemotherapeutic agent. Enzalutamide blocks AR nuclear translocation and DNA binding, suppressing AR-driven gene expression. Paclitaxel inhibits cell division by stabilizing microtubules and preventing their depolymerization. This combination has been studied primarily in hormone receptor-positive cancers such as luminal androgen receptor (LAR)-enriched triple-negative breast cancer (TNBC), where AR signaling may drive tumor growth. The rationale for combining these agents is based on potential synergistic antitumor effects observed in preclinical models and early-phase clinical trials[1][2][4]. Clinical studies have shown that this regimen can be effective and safe as neoadjuvant or advanced therapy for TNBC patients with high AR expression[1][2][7].
