Overview

Enzelkitug (RG6424) is a recombinant human interleukin-22 (IL-22) Fc-fusion protein developed by Genentech, a subsidiary of Roche. It consists of the human IL-22 cytokine sequence fused to the Fc region of a human IgG1 immunoglobulin. Enzelkitug functions as an agonist of the IL-22 receptor complex, which is composed of the IL-22R1 and IL-10R2 subunits. While IL-22 is traditionally recognized for its role in promoting epithelial regeneration and mucosal barrier integrity—leading to its investigation in inflammatory bowel disease—it is also being evaluated in oncology. In the context of advanced solid tumors, enzelkitug is being studied both as a monotherapy and in combination with immune checkpoint inhibitors (such as atezolizumab or pembrolizumab) to determine its safety, pharmacokinetics, and potential to modulate the tumor microenvironment to enhance anti-tumor immune responses.
