EO-3021 + ramucirumab

EO-3021 + ramucirumab is a combination of two investigational and approved oncology drugs currently being evaluated for the treatment of advanced or metastatic gastric or gastroesophageal junction (GEJ) cancer expressing Claudin 18.2. EO-3021 is an antibody-drug conjugate (ADC) targeting Claudin 18.2, developed by Elevation Oncology, intended to deliver cytotoxic payload specifically to tumor cells expressing this antigen. Ramucirumab is a human IgG1 monoclonal antibody (approved as Cyramza) targeting vascular endothelial growth factor receptor 2 (VEGFR2), inhibiting ligand-stimulated angiogenesis in tumors. The combination aims to provide potentially enhanced antitumor efficacy due to the distinct and complementary mechanisms of action—direct tumor targeting/cell killing and inhibition of tumor angiogenesis. Preclinical studies have shown superior tumor growth inhibition with the combination compared to either agent alone. The combination is under evaluation in a Phase 1 clinical trial in the second-line treatment setting for gastric and gastroesophageal junction cancers[1][3][5][7].
