Overview

EO2401 is an off-the-shelf, first-in-class peptide-based cancer immunotherapy developed by Enterome. It is designed as a therapeutic vaccine to activate and expand memory T cells that cross-react with tumor-associated antigens (TAAs) present on certain cancers. The vaccine contains three synthetically produced HLA-A2-restricted peptides (OncoMimics) derived from gut microbiome proteins that mimic TAAs upregulated in glioblastoma—specifically IL13Rα2, BIRC5 (survivin), and FOXM1—as well as a CD4 helper peptide UCP2. Upon administration, these peptides are presented by dendritic cells to T cells, inducing a cytotoxic immune response against tumor cells expressing the corresponding TAAs. EO2401 has been studied primarily in combination with immune checkpoint inhibitors such as nivolumab ± bevacizumab for recurrent glioblastoma and other solid tumors including adrenocortical carcinoma and pheochromocytoma/paraganglioma[3][5][6][8]. EO2401 leverages molecular mimicry between microbial peptides and human tumor antigens to stimulate anti-tumor immunity. It is not FDA approved or commercially available; it remains investigational[7]. The mechanism of action involves immunostimulation via activation of effector memory CD8+ T cells targeting specific TAAs[3][6]. Clinical trials have shown promising immune responses and safety when combined with checkpoint inhibitors in recurrent glioblastoma patients.
