Overview

EO2401 + nivolumab is a combination therapy composed of EO2401, a microbiome-derived peptide vaccine designed to induce a CD8+ T-cell immune response against tumor antigens, and nivolumab, a monoclonal antibody immune checkpoint inhibitor targeting PD-1. EO2401 contains three synthetic peptides that mimic epitopes from tumor-associated antigens IL-13Rα2, BIRC5/survivin, and FOXM1, along with a helper peptide (UCP2); it is administered subcutaneously with the adjuvant Montanide. Nivolumab blocks PD-1 on T cells to enhance antitumor immune activity. The combination aims to harness and potentiate immune responses in cancers such as recurrent glioblastoma and adrenal malignancies. EO2401 is developed by Enterome, while nivolumab is developed and marketed by Bristol Myers Squibb. Clinical trials (e.g., ROSALIE, Spencer) highlight immune activation, manageable safety profile, and encouraging preliminary efficacy in these diseases[1][2][3][4][5].
