Overview

EO4010 is an investigational, off-the-shelf **multi-peptide therapeutic cancer vaccine** developed by Enterome using its OncoMimics platform. It comprises five synthetic microbial-derived peptides designed to mimic HLA-A2-restricted CD8-positive T-cell epitopes from the tumor-associated antigens survivin, forkhead box protein M1, ubiquitin-conjugating enzyme E2 C, cell division cycle protein 20, and kinesin-like protein KIF2C, plus the CD4-positive helper peptide universal cancer peptide 2. By presenting non-self bacterial peptide mimics, EO4010 is intended to expand pre-existing memory T cells that cross-react with the corresponding tumor antigens, thereby promoting targeted cytotoxic immune responses against tumor cells. It was evaluated in the Phase 1/2 AUDREY study in previously treated, unresectable microsatellite-stable or mismatch-repair-proficient metastatic colorectal cancer, as monotherapy and with nivolumab with or without bevacizumab.
