eOD-GT8 60mer is an experimental HIV-1 vaccine candidate designed using a germline-targeting strategy. Developed by the International AIDS Vaccine Initiative (IAVI) in collaboration with Scripps Research, it consists of an engineered outer domain (eOD) of the HIV-1 gp120 protein. This immunogen is specifically designed to have high affinity for the germline precursors of VRC01-class broadly neutralizing antibodies (bnAbs). The eOD is linked to a lumazine synthase (LumSyn) protein, which self-assembles into a 60-mer icosahedral nanoparticle to enhance immune recognition. In clinical trials, it is typically administered with the AS01B adjuvant. The primary goal of the vaccine is to prime rare B cell precursors that can eventually be matured into bnAbs through subsequent booster immunizations to prevent HIV infection.
