Overview

EOL-101 (also known as EORA-101) is an investigational small molecule orexin-1 receptor (OX1R) antagonist being developed by Eolas Therapeutics. It is designed to target neural pathways within the brain's reward system to treat substance use disorders, with a primary focus on nicotine addiction and smoking cessation. By selectively blocking the downstream OX1R localized in the hypothalamus, EOL-101 aims to directly modulate the motivational properties of addiction and prevent cue-induced relapse without causing the side effects associated with upstream nicotine receptor blockers or dual orexin receptor antagonists.
