eOPN3 is a red-shifted, inhibitory type-2 opsin derived from encephalopsin (OPN3), a G protein-coupled receptor (GPCR). It is utilized as an optogenetic tool for precise neuromodulation. When expressed in neurons (often via viral vectors like AAV) and activated by light, eOPN3 triggers the Gi/o signaling pathway, leading to the suppression of synaptic transmission and neuronal activity. This inhibitory effect is being investigated for the treatment of drug-resistant focal epilepsies, such as mesial temporal lobe epilepsy (mTLE), by modulating complex epilepsy networks like the hippocampus-anterior nucleus of the thalamus (HPC-ANT) circuit.
