Overview

EP-027367 is a small molecule core inhibitor, also known as a capsid assembly modulator (CAM), developed by Enanta Pharmaceuticals for the treatment of chronic hepatitis B virus (HBV) infection. It specifically targets the HBV core protein (capsid), disrupting the viral lifecycle at multiple stages, including the assembly of viral capsids and the replication of viral DNA. In preclinical studies, including chimeric mouse models with human liver cells, EP-027367 demonstrated potent antiviral activity, achieving up to a 3.0 log reduction in HBV DNA levels from baseline. The compound exhibits pan-genotypic activity and has shown the potential to prevent the establishment of covalently closed circular DNA (cccDNA) in vitro, which is a critical factor in achieving a functional cure for HBV.
